93 g propionyl chloride
133 g anhydrous AlCl₃
Solvent: carbon disulfide or DCM
Reflux 2-3 hours after adding slowly at 0-5°C.
Hydrolyze, extract, distill.
Yield: 70-85%
2)-Alpha-bromination
Ketone + Br₂ in glacial acetic acid or chloroform
2-bromo-1-(thiophen-2-yl)propan-1-one
Bromine adds to alpha carbon, NOT the ring

3)=Amination
Bromoketone + excess methylamine (40% aq or gas in methanol) → imine
Reduce immediately:
NaBH4/methanol
or
Al/Hg amalgam (classic method)
→ racemic methiopropamine freebase
Alternative: direct reductive amination of the ketone with MeNH2 + NaBH3CN or NaBH(OAc)3/methanol.
Workup & salt
Extract base with non-polar solvent; dry; bubble dry HCl gas or add HCl/IPA → hydrochloride salt (white brittle crystals; hygroscopic; mp 170–180 °C).

Route 2 – Nitropropene route (dirtier, but easier precursors)
Thiophene-2-carbaldehyde + nitroethane + catalyst (ammonium acetate or n-butylamine) → 1-(thiophen-2-yl)-2-nitroprop-1-ene (Henry/Knoevenagel condensation)
Reduce the nitroalkene
Options:
Fe/HCl or Zn/acetic acid → primary amine (then N-methylate with MeI/base or formaldehyde + NaBH₃CN)
LiAlH₄ in ether/THF → primary amine directly
